Our research plan is to profile the expression of glycans in hES cells during undifferentiated and differentiated stages. The primary aim of the study is to characterize known and novel glycans to both investigate their function and correlate their expression with cellular phenotype. Initial experiments will include (1) identification of glycoepitopes using lectin arrays, (2) metabolic incorporation of unnatural monosaccahrides onto the cell surface, (3) tracing of known markers using antibody labeling, and (4) the use of glycosyltransferase inhibitors as well as measurement of glycosyltransferase expression during various stages of hES cell differentiation.
